Not particularly. The game just uses these names as a reference to make the bosses appear.

They actually do have internal names, Pixel just didn't make them very obvious or easy to find. For my first playthrough it took me maybe 3 or 4 hours, NG+ took me a couple hours more. The game gets easier each playthrough (you get used to the stages), and combined time for the game and NG+ on my second playthrough was only 3 and a half hours. It's quite a challenging game though.
