They have lines on their faces in prerelease art and the Nicalis art, but not in-game (even in the facepics and credits)

I think he means the "Tear" lines coming from the eyes

Yeah, those lines are also there in all the Cave Story 3D promotional art (at least the North American version).

I've always preferred Quote without those lines descending from his eyes that look like the trajectory of tears. They seem to be meant to make it more obvious that he's a robot, but it's not an artistic move that really appeals to me. I also don't particularly like the depictions of Quote that give him really weird body proportions (like his model in Cave Story 3D), and I also don't really like the art that depicts him as a teenager or adult. To me, the definitive art showing Quote would have to be the credit art images.

And in my opinion, the most accurate promotional art was this picture which I believe was promotional art by NICALiS for Wiiware port back in 2009:

I agree 100%. This was the first time I ever saw Quote- it was in a game informer magazine which reviewed the wii ware version of cave story. When I first saw this picture I honestly thought quote was a girl because of his appearance. (But then again, I was 12- give or take, and couldn’t judge so good.)

I think it's to add the distinction between human & robot. I usually draw quote with thr face lines, but not curly.

The tear lines are really bad. The fanart utilizing the tear lines is even worse.

I've never been much of a fan of them. It works in some of Pixel's old artwork, e.g. like

but any more than that and it gets distracting, especially if they're drawn to be more realistically artificial which instead makes them look like creepy doll people.
